Chromatomics
a) Purification of Chromatin Domains
We study the total protein composition of chromatin fibers that contain specific proteins such as HP1, histone variants or carry particular histone modifications. We do this by shotgun proteomics and label free quantitation.
b) Histone Modification Spectral Library
In order to facilitate and streamline histone analysis using mass spectrometry we will generate a high quality spectral library and have generated a set of isotopically labeled standard peptides that increase the speed and the accuracy of modification profiling.
c) Impacts of Nutrition on Epigenetic states
We have identified a histone methyltransferases that is very sensitive towards changes in media composition and intracellular SAM levels. As we detect the SAM synthetase (SAMs) in the nucleus of Drosophila tissue culture cells we will study the protein-protein interactions and the interactions of SAMs with chromatin within the Epigenesys network.
