The Almouzni lab's major scientific objective will be to gain data enabling researchers to model nucleosome assembly with distinct variants and organisation of heterochromatin domains, to assess their plasticity/stability throughout the cell cycle and when challenged under various perturbations. For this, the Almouzni lab will use biochemical and cellular assays involving protein complexes (such as recombinant histone chaperones, histone H3 variants, HP1, epitope tagged in vivo) to study the dynamics and modifications of histone H3 variants and heterochromatin proteins during the cell cycle.
